Antoine was the uncle of Mary of Guise.

In 1527, Antoine's son became engaged to Anne of Cleves, but as she was only 11 and the prospective groom only 10, the betrothal was not considered official and was later called off.

At court as a young man, Antoine became good friends with Francis I of France, before he was king.

Antoine would have been familiar with interacting with King Louis XII of France as a courtier, and in 1509, he travelled to northern Italy into battle with him

Despite his friendship with the king, Antoine refused to take sides in Francis I's war with his cousin Charles V, Holy Roman Emperor. After the Treaty of Nuremberg in 1542, Charles V granted Antoine's duchy of Lorraine independence as a gesture of gratitude for his sympathy.

Antoine was painted by Hans Holbein the Younger in 1543.
